To measure proline by spectrophotometric method with the formation of a
color complex by ninhydrine, solutions with a variety of standard
concentrations of proline have been prepared and the standard graph for it has
been drawn. In this method, the lowest concentration of solution is equal to 2
.ppm
Surfactants can increase dynamic range, decrease detection limit and improve
correlation coefficient of benchmarking graphs by creating Mycelian medium.
In this research, the effects of cationic and anionic and non-ionic surfactants
have been investigated and it is observed that non-ionic acid oleic surfactant
increases molar absorption coefficient by creating a Mycelial medium. The
critical concentration of mycellation was obtained for this surfactant and after
optimizing other conditions, it was possible to measure proline even with a
.concentration of 300 ppb
After drawing the benchmarking charts, a positive effect was observed in
increasing dynamic range, reducing detection limit and improving the
correlation coefficient.
